You know, those small plastic dispensers in various shapes and colors that you load with the tiny candy that became a big hit in the Fifties and Sixties. Who hasn't had one of those growing up But talk about taking it to the next level! When Glew learned that the 'Pezident' of Pez U. S. A. Nixed a whole variety of dispensers from the world headquarters manufacturing facility in Austria, he got the bright idea of traveling to Europe to bring home satchels full of the things to sell to a fan base willing to shell out big bucks for the rarities. Eventually becoming a big name among the Pez collecting community, Glew drew the attention of Scott McWhinnie, President of Pez U. S. A., who made it a personal vendetta to put an end to Glew's quasi-illegal operation. Maybe he lost a bet. Maybe he was bored from his day off from Criminal Minds and wanted to watch a naked stripper being hunted down in the desert. The pop ending was a surprise, but not enough to even earn a second star. This was a pretty decent thriller/survival horror movie. A young woman in a small town in New Mexico forced to work as a stripper, is taken by a local hunter and brought naked into the wilderness in order for him to hunt her down and kill her. This was pretty well made, the writing was basic, but the intensity of the hunt was crafted well. An unexpected gem. And Starring: Joe Mantegna gets a rather prominent credit at the end of the sequence. Beauty Is Never Tarnished: Despite being fully exposed in the wilderness for a day and a half, the worst Diana has to do is wrap her feet in cloth to protect them from the ground. Bittersweet Ending: Diana manages to kill Colin and return to town, but her ordeal has left her distrustful of everyone, and she escapes into the night. Ten months later, she's back on the prowl killing sex offenders and becomes known as the \"Southwest Slayer\". Creepy Souvenir: Colin keeps a collection of jewelry from the women he kills. In the opener, he takes a bracelet from his most recent victim and later adds it to a collection of many other jewelry pieces. Cruel and Unusual Death: Being hunted down while naked, and then being unceremoniously buried with no one else knowing just what happened to you, is an extremely undignified way to go. Gory Discretion Shot: There is only one time a shooting occurs on-screen, and even then it's far away, so you're spared the grisly details. Junkie stripper tells her all the ways she can make more money- take off more clothes, interact with patrons, and offer other services. At first Diana isn't interested but she needs the money so she gives it a try. It yields her more money and the night she meets a patron and drives away with him she ends up changing her mind about offering him other services. But he won't have none of it. He ends up kidnapping her.Next, she wakes up out in the desert nude...as prey for our hunter. He gives her a 15 minute head start. He catches up with her but he likes a challenge so he doesn't make it that easy for himself. And Diana, is also a bright girl. She tricks him and escapes from him repeatedly, bloody and bruised. Eventually she gets the upper hand and runs into a family camping, the dad and his two teenage sons, who are stunned to see a mostly naked girl who now is in shock. Dad does the right thing and leaves to get help. But the hunter isn't done yet. Nor is Diana.Naked Fear is an excellent movie for what it is- a well-done B movie with a great story and good acting. Of course seeing the lovely Danielle De Luca run around naked for a good while doesn't hurt a bit. Her performance overall is remarkable, downright award-worthy, given what she has to go through. It obviously suffers from B-movie weaknesses- low budget, not so good acting by secondary characters, an unpolished look indoors, etc. It's a tad bit too long, certain things could have been cut out or shortened. At the same time another victim should have been added, just to make our villain more despicable.The movie succeeds at presenting the hardships of small town life and the risks of trying to make it elsewhere pretty well. And it does so without being condescending and offensive. You do end up empathizing with the main character. The movie is never exploitative but manages to humanize our characters and give the story a lot of meaning. Emphasis was given to the comparison of the thermal and optical performance between the designs, as well as in the temperature levels of the fluids and the receiver. The geometry of the integrated cavity receiver was optimized according to two independent parameters and two possible optimum designs were finally revealed. The optimization took place regarding the optical performance of the collector with the cavity receiver. The simulation results indicated that the cavity design leads to enhancements of up to 4.40% and 4.00% in the optical and thermal efficiency respectively, while the minimum possible enhancement was above 2.20%. The mean enhancements in optical and thermal performance were found to be 2.90% and 2.92% respectively. Moreover, an analytical solution was developed for verifying the numerical results and the maximum deviations were found to be less than 5% in all the compared parameters. Especially, in thermal efficiency verification, the maximum deviation took a value of less than 0.5%. In line with the free drug hypothesis, the use of unbound brain concentrations (Cu,br) has been shown to provide the best correlations with pharmacological data. However, typically the determination of this parameter requires microdialysis, a technique not ideally suited for screening in early drug development. The main objective of this study was to explore the possibility to directly generate Cu,br/Cu,pl ratios in a single in vitro model of the BBB, using a co-culture of brain capillary endothelial and glial cells in an attempt to mimick the in vivo situation, thereby greatly simplifying existing experimental procedures. Comparison to microdialysis brain concentration profiles demonstrates the possibility to estimate brain exposure over time in the BBB model. A stronger correlation was found between in vitro Cu,br/Cu,pl ratios and in vivo Cu,br/Cu,pl obtained using fu,br from brain slice than with fu,br from brain homogenate for a set of 30 drugs. Overall, Cu,br/Cu,pl ratios were successfully predicted in vitro for 88% of the 92 studied compounds. This result supports the possibility to use this methodology for identifying compounds with a desirable in vivo response in the CNS early on in the drug discovery process. The main objectives of this study were to (i) compare the in vitro Cu,br/Cu,pl ratios generated using the BBB model with in vivo brain concentration profiles from microdialysis experiments. Rat plasma was obtained from Sprague-Dawley rats in-house. Plasma from three individuals was pooled and mixed with up to 4 compounds with a concentration of 10 µM each (in 0.1% DMSO). A 48-well dialysis apparatus (AstraZeneca, Mölndal) was used to determine the plasma free fraction. The dialysis membrane (AstraZeneca, Mölndal) was conditioned in Milli/Q water before use. A volume of 180 µl 0.122 M phosphate buffer, pH 7.4 with 75 mM NaCl constituted the receiver solution and 180 µl of the plasma/compound mixture were added to the donor side. The samples were incubated on an orbital shaker (4 mm in diameter and 100 rpm) at 37oC for 18 hrs. Following incubation, 50 µl aliquots were removed from the respective plasma and buffer sides from triplicate equilibrium dialysis chambers and transferred to a 96-well assay block (Costar Brand 96-well assay blocks, polypropylene (sterile, 0.5 ml, V-bottom) (Fisher Cat No. 07200723)). After plasma protein precipitation by the addition of 150 µl of acetonitrile to each plasma sample in order to precipitate plasma proteins. The plates were centrifuged and the supernatant analyzed by LC-MS/MS. The fu in plasma was calculated from the ratio of the MS-area of the compound in the buffer to the MS-area of the compound in the plasma.
